A Randomised, Double-Blind, Placebo-Controlled, Cross-Over Clinical Trial to Evaluate the Biological Effects and Safety of a Polyphenol Supplement on Healthy Ageing

DailyColors™ is a supplement containing various phytonutrients designed to mimic aspects of the Mediterranean diet. It includes fruit, berry, and vegetable extracts rich in vital phytochemicals like Quercetin, Catechins, Phloretin, Ellagic Acid, and Anthocyanins. This study by Chong et al. (2024) aimed to assess the impact of DailyColors™ on blood biomarkers related to aging and age-related diseases, particularly those involving mitochondrial function, inflammation, oxidative stress, and saliva DNA methylation patterns. Thirty adult participants (average age 67.0 years, SD = 7.5) with a body mass index over 25 were enrolled in this randomized, double-blind, placebo-controlled, cross-over trial, which consisted of two one-week treatment periods, separated by a one-week washout period. Blood CD38 concentrations significantly increased from baseline to 24 hours during the placebo phase, a change not seen during the active phase. Elevated CD38 is linked to mitochondrial dysfunction and inflammation. Additionally, plasma levels of 4-HNE, a marker of oxidative stress, showed a downward trend after a week of DailyColors™ supplementation. After a one-month open-label follow-up with 26 participants, the authors detected hypermethylation of the CpG site cg13108341, reversing the trend typically seen in aging. In conclusion, while the effects were minimal, DailyColors™ supplementation may help address age-related changes.
